ABSTRACT

The objective of this study was to analyze whether obese women with no metabolic syndrome (MetS) have increased cardiometabolic risk compared to non-obese women and to observe the correlations between adiposity and coronary heart disease (CHD) risk factors in metabolically healthy women. 20-40 year old non-obese (n=41), obese with no MetS (n=30) and obese with MetS (n=28) women were studied. Lipid profile, blood pressure, CHD family history, physical inactivity, high-sensitivity C-reactive protein (hs-CRP), fibrinogen, interleukin-1β and tumor necrosis factor-alpha were analyzed. A subset of obese (13) and non-obese (33) women with no major components of MetS (except waist circumference) were further compared. Obese women with no MetS and non-obese women presented a similar metabolic profile that was statistically different from those seen in obese women with MetS. The number of obese women with no MetS and non-obese women presenting two or more risk factors (23.3 and 19.5%, respectively) or presenting high Framingham Risk Score (6.7 and 2.4%, respectively) were also similar. The only pro inflammatory protein correlated to waist circumference was hs-CRP. These data suggest that obesity with no MetS induce a CHD risk comparable to the risk seen in non-obese women. However, when women with no major components of MetS alone were considered, adiposity was positively correlated to blood pressure and hs-CRP. Although CHD risk of obese women with no MetS is closer to non-obese women, adipose tissue expansion was positively correlated to blood pressure and hs-CRP that are important risk factors for CHD.


Obesidad sin síndrome metabólico y expansión del tejido lipidio basado exclusivamente en factores de riesgo e marcador inflamatorio de la enfermedad coronaria em mujeres en la pré-menopausia. El objetivo de esta investigación fue analizar si las mujeres obesas que no tienen el síndrome metabólico (MetS), tienen riesgo cardiometabólico aumentado comparado con mujeres no obesas y observar las correlaciones entre factores de riesgo de la adiposidad y la enfermedad coronaria del corazón (CHD) en mujeres metabólicamente saludables. Fueron estudiadas mujeres de 20-40 años de edad no-obesas (n=41), obesas sin MetS (n=30) y obesas con MetS (n=28). Se analizaron también factores tradicionales de riesgo y marcadores inflamatorios. Un sub conjunto de mujeres obesas (13) y no obesas (33) sin componentes mayores de MetS fueron comparados adicionalmente. Mujeres obesas sin MetS y mujeres no obesas evidenciaron un perfil metabólico semejante, estadísticamente diferente de lo visto en mujeres obesas con MetS. El número de mujeres obesas sin MetS y no obesas que presentaban dos ó más factores de riesgo (23.3 y 19.5%, respectivamente) ó presentaban riesgo alto con Framingham (6.7 y 2.4%, respectivamente) también fueron semejantes. Estos datos sugieren que la obesidad sin MetS induce un riesgo de CHD comparable al riesgo observado en mujeres no obesas. Sin embargo, cuando las mujeres sin componentes importantes del MetS únicamente fueron llevadas en cuenta, la adiposidad fue correlacionada a la presión sanguínea y a la hs-CRP. Aunque el riesgo de la CHD de mujeres obesas sin MetS sea más próximo al de mujeres no obesas, la expansión del tejido adiposo fue positivamente correlacionado a la presión sanguínea y a la hs-CRP, ambos importantes factores de riesgo para la CHD.

ABSTRACT

OBJECTIVE: The objective was to assess the effects of oral administration of sodium butyrate on colon carcinogenesis. METHODS: Carcinogenesis in adult male Wistar rats was induced with 1.2-dimethylhydrazine injections at a dose of 40mg/kg of body weight. A solution of sodium butyrate (3.4 percent) was given ad libitum for 4 weeks (butyrate group, n=16) instead of water (control group, n=9). Rats were killed 17 weeks after 1.2-dimethylhydrazine administration. Aberrant crypt foci and expression of the messenger ribonucleic acid (mRNA) of cyclins D1 and E were quantified in the colon. Alterations in the fatty acid profile of the colon, liver, intra-abdominal fat and feces were also analyzed. RESULTS: A significant decrease in aberrant crypt foci was found in the group taking butyrate. No differences were found between the groups in the mRNA expression of cyclins D1 and E. Nevertheless, butyrate intake decreased the content of stearic and oleic acids in the intra-abdominal fat and docosahexaenoic acid in the liver. Moreover, these rats presented higher percentages of linoleic acid in the intra-abdominal fat than control rats. CONCLUSION: The data indicate that butyrate use in rats reduced preneoplastic lesions and changes in the intra-abdominal fat and fatty acid profile of the liver, commonly found in colon carcinogenesis.


OBJETIVO: Avaliar o efeito da administração oral de butirato de sódio na carcinogênese do cólon. MÉTODOS: A carcinogênese em ratos Wistar foi induzida com injeções de 1,2-dimetilhidrazina na dose de 40mg/kg de peso corporal. A solução de butirato de sódio (3,4 por cento) foi oferecida ad libitum por 4 semanas (grupo butirato, n=16), em substituição à água (grupo controle, n=9). Os ratos foram sacrificados na 17ª semana após tratamento com a 1,2-dimetilhidrazina. Focos de criptas aberrantes e a expressão dos genes para o ácido ribonucléico mensageiro (RNAm) das ciclinas D1 e E foram quantificadas no cólon. Alterações no perfil de ácidos graxos do cólon, no fígado, na gordura intra-abdominal e nas fezes foram analisadas. RESULTADOS: Uma significante diminuição nos focos de criptas aberrantes foi encontrada no grupo que recebeu butirato. Nenhuma diferença foi encontrada na expressão do RNAm das ciclinas D1 e E entre os grupos. Contudo, a ingestão de butirato diminuiu a quantidade de ácido esteárico e ácido oléico na gordura intra-abdominal e do ácido docosahexanóico no fígado. Além disso, o grupo butirato apresentou maior percentual de ácido linoléico na gordura intra-abdominal, comparado com os ratos do grupo controle. CONCLUSÃO: Os dados indicam que o uso de butirato reduz lesões pré-neoplásicas em ratos e diminui as alterações no perfil de ácidos graxos da gordura intra-abdominal e do fígado, comumente encontradas na carcinogênese colônica.

ABSTRACT

OBJECTIVE: Surgical scar tensile strength may be influenced by several factors such as drugs, hormones and diet. The purpose of the present study was to determine the influence of a shrimp-enriched diet on the tensile strength of rat scars. METHODS: Forty male Wistar rats were submitted to a 4 cm dorsal skin incision and the wounds were sutured with 5-0 nylon interrupted suture. The animals were divided into two groups: Group 1 (control) received a regular diet, and Group 2 (experimental) received a shrimp-enriched diet. The two diets contained the same amounts of proteins, lipids and carbohydrates. The rats in each group were divided into two subgroups according to the time of assessment of the scar tensile strength: subgroup A, studied on the 5th postoperative day, and subgroup B, studied on the 21st postoperative day. RESULTS: The tensile strength of the scar on the 5th postoperative day was lower in the animals that received the shrimp-enriched-diet (303.0, standard error of mean= 34.1) than in the control group (460.1, SEM = 56.7) (p<0.05). CONCLUSION: A shrimp diet reduces the tensile strength of the scar. The next step of this study will be to clarify the mechanism in which shrimp affects tensile strength.


OBJETIVO: A resistência cicatricial da pele pode ser influenciada por diversos fatores como medicamentos, hormônios e dieta. Este trabalho foi delineado para determinar a influência da dieta com camarão na resistência cicatricial na pele. MÉTODOS: Quarenta ratos machos Wistar foram submetidos a incisão (4cm) e suturas interrompidas da pele dorsal, com fio de nylon 5-0, e foram divididos em dois grupos: o Grupo 1 (controle) recebeu uma dieta convencional e Grupo 2 (experimental), recebeu dieta com adição de com camarão. As duas dietas continham quantidades semelhantes de proteína, lipídeos, e carboidratos. Os ratos de cada grupo foram divididos em dois subgrupos de acordo com os distintos períodos pós-operatórios de avaliação da resistência tecidual: subgrupo A, estudado no 5° dia pós-operatório, e subgrupo B, estudado no 21° dia pós-operatório. RESULTADOS: A resistência cicatricial da pele no 5° dia pós-operatório foi menor nos animais que receberam dieta suplementada com camarão (303,0, erro padrão da média=34,1), quando comparada ao grupo controle (460,1, erro padrão=56,7) (p<0,05). CONCLUSÃO: A dieta suplementada com camarão reduziu a resistência cicatricial da pele de ratos. Dando continuidade ao estudo, será averiguado o mecanismo pelo qual ocorre essa redução.
